Abstract
Introduction: The precuneus(Pc)isacentralhubintegratingdiversebrainfunctions,anditisearlyinvolvedinAlzheimer’s
disease (AD)pathophysiology,yetitssubregionalvulnerabilityremainspoorlyunderstood.Thisstudyaimstodelineatethe
diBerential involvementandvulnerabilityofthesesubdivisionsintheearlystagesofAD.
Methods: We conductedaresting-stateFCanalysisofPcsubregionsin32cognitivelyunimpairedcarriersofthePSEN1E280A
mutation comparedto25noncarriersusingseed-basedanalyses.
Results: As comparedtononcarriers,wefoundthatthegroupofasymptoticcarriershadincreasedconnectivityfromthe7Am
subregion towardtheentorhinalcortex,superiortemporalgyrus,insula-operculum,dlPFC,andsomatosensoryareas.Moreover,
the POS2subregionsigniFcantlydecreasedconnectivitywiththeanteriorinsulaanddlPFC.HigherMoCAscorescorrelatedwith
increased connectivitywithinandbetweenthePcandfrontoparietalnetworks.
Discussion: These resultsrevealdistinctpatternsofearlyfunctionalreorganizationandsupportthesubregionalPcanalysisin
